membrane labeling

SNAP
  • {myrSNAP}attP40 or {myrSNAP}attP2 better signal in neurites
  • CD4-SNAP/CyO;CD4-SNAP/TM6b stock enriched in cell bodies
CLIP
  • CD4-CLIP#1/CyO no myrCLIP available at the moment
Halo
  • have used single attP40 and attP2 insertions which all worked; could try two insertions
TMP
  • {myrTMP}attP40/CyO; {myrTMP}attP2 even when using this double insertion, signal is very weak (increase fixation to 40-60 min); only really useful for neuropil labeling

synaptic labeling

only SNAP and CLIP at the moment; in general, SNAP gives better signal-to-noise

pre

  • SytCLIP#4/CyO or SytCLIP#6/TM3 these two P-element insertions have high expression levels (see list)
  • SytSNAP#3/CyO or SytSNAP#2/TM3 these two P-element insertions have high expression levels (see list)
  • CLIPsyb P-element insertions on 2 and 3, haven't thoroughly compared expression levels
  • SNAPsyb P-element insertions on 2 and 3, haven't thoroughly compared expression levels

post

  • TLN-CLIP#6/CyO or TLN-CLIP#9/TM3 these two P-element insertions have high expression levels (see list)
  • TLN-SNAP#4/CyO or TLM-SNAP#5/TM3 these two P-element insertions have high expression levels (see list)

neuropil labeling

  • brp-SNAP/CyO knock-in based on MiMIC insertion
  • dlg-SNAP knock-in based on MiMIC insertion
  • dlg1-SNAP product of P-element swap of protein trap line; weaker than the knock-in line
  • dlg1-CLIP product of P-element swap of protein trap line

General Performance of tags

  • labeling speed: SNAP > Halo > CLIP > TMP
  • signal-to-noise: Halo > SNAP > CLIP > TMP
  • signal intensity (with best available subtrate): SNAP,Halo > CLIP > TMP

take into consideration e.g. when using one tag for single-cell labeling and a second one for neuropil labeling
