  1. I am having trouble opening the images.
    • All images can be opened by the stock version of Fiji. You can then convert to pretty much any format you want. See also Software page.
  2. In the Successfully Registered image, some of the images have three channels. I am wondering what is the '03' stained for?
    • this should be UAS:Synaptotagmin-HA stained using anti-HA. See Supplemental Experimental Procedures for further details.
  3. I have another question about the correspondence between the image files. For example, is the 'aTD-a_mask_SAEJ26-1.nrrd' in the ExtractedImages corresponding to the 'IS2_SAEJ26-1_01_warp_m0g80c8e1e-1x26r4.nrrd' and 'IS2_SAEJ26-1_02_warp_m0g80c8e1e-1x26r4.nrrd' in the Successfully Registered image, or SAEJ26 is the marker for the same images in all the folders on the disk?
  4. You've given me a bare hard drive. What should I buy to use it?
    • You need a 3.5" SATA enclosure. You can buy them for <$40 (or just ask you IT dept). Make sure cables are included esp eSATA or FW. Best interface depends on your platform.
    • Mac: Firewire 800 for good performance on all recent macs (400 for pre 2007? machine)
    • PC: eSATA for excellent performance on new PCs
    • USB2.0 slow, but very cheap and works on all machines
